Mode of Action.

The GABA A receptor complex includes binding sites for benzodiazepines, barbiturates, and steroids. When the neurotransmitter GABA binds with the receptor it triggers a conformational change in the pore, allowing more Cl- to enter the cell (Nutt & Malizia, 2001). Increased GABA binding increases the frequency of opening the associated Cl- channel, consequently amplifying the inhibitory effect (Cooper, Bloom, & Roth, 1996).

Zolpidem also demonstrates selectivity for GABA A receptors containing α 1 subunits. The new generation hypnotic drugs, zolpidem, zopiclone and zaleplon, are at least as efficacious in the clinic as benzodiazepines and may offer advantages in terms of safety. Of particular interest is the finding that zolpidem shows a wide separation between doses producing sedative effects and those giving rise to other behavioural actions, and induces less tolerance and dependence than benzodiazepines. Recent studies using genetically modified mice have confirmed that receptors containing alpha 1 subunits play a particularly important role in mediating sedative activity, thus providing an explanation for the pharmacological profile of zolpidem. These drugs act through the BZ binding sites associated with GABA A receptors, but show some differences from benzodiazepines in pharmacological effects and mechanisms of action.

Although it has been known for quite some time that BZs modulate GABA receptors, the molecular mechanism is still unresolved. ? ?. The binding of GABA to postsynaptic GABA receptors opens a chloride-selective ion pore that is an integral component of the multimeric receptor complex. Furthermore, GABA receptors are a primary target for several neuroactive drugs including barbiturates, steroids, general anesthetics, and benzodiazepines (BZ); the latter of which is the subject of this proposal. Gamma aminobutyric acid (GABA) is the major inhibitory neurotransmitter in the mammalian brain. The resulting chloride flux across the cell membrane inhibits the postsynaptic neuron. This proposal will use a combination of molecular biology, electrophysiology, single oocyte radioactive ligand binding, and site directed fluorescent labeling to gain structural and functional insights into the actions of BZs on GABA receptors. Some of these techniques are new to the BZ field and it is hoped they will bring some fresh perspectives to the problem. The results from these proposed studies are expected to contribute to that effort. ? The design of more efficacious BZs that can target the many different GABA receptor subtypes that have been identified in the brain will ultimay depend on understanding the structural requirements and precise mechanism of action of this important class of neuroactive compounds. Dysfunctions in GABA-mediated inhibition have been implicated in the etiology of a variety of neurological and psychological disorders.
